Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
Tirzepatide is a synthetic peptide that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. The molecule contains 39 amino acids and features a C20 fatty diacid moiety attached via a linker, which promotes albumin binding and extends its circulating half-life. Its sequence incorporates non-natural amino acids and modifications that reduce susceptibility to degradation by dipeptidyl peptidase-4. This dual receptor activity distinguishes it from selective GLP-1 receptor agonists.
The GIP receptor is expressed in pancreatic islets, adipose tissue, and the central nervous system, while GLP-1 receptors are found in pancreatic islets, the gastrointestinal tract, and the brain. Activation of both receptors can enhance glucose-dependent insulin secretion and reduce glucagon release. The relative contribution of each receptor to the overall pharmacological effect remains an area of ongoing investigation. Preclinical studies suggest that GIP receptor agonism may modulate appetite and energy balance, but the precise mechanisms in humans are not fully established.

Routine characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity assessment, usually with ultraviolet detection near 214 nanometers. Intact mass measurement by liquid chromatography coupled to mass spectrometry confirms molecular identity against a theoretical value. Sequence-level confirmation uses enzymatic digestion followed by tandem mass spectrometry, an approach known as peptide mapping. Amino acid analysis gives an independent check on composition. Circular dichroism spectra are used to estimate helical content in aqueous buffer.
Stability depends strongly on physical form. The dry powder is generally regarded as stable for extended periods when held at or below minus twenty degrees Celsius in a sealed, desiccated container. In solution, degradation pathways include deamidation of asparagine and glutamine residues, oxidation of methionine, and aggregation. Reaction rates for these pathways rise with temperature. Repeated freezing and thawing of solutions promotes aggregation, and light exposure can accelerate some oxidative changes. Buffer composition and pH influence which pathway dominates at a given temperature.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.
=== Biosynthetic pathway === Plants exclusively of the genus Capsicum produce capsaicinoids, which are alkaloids. Capsaicin is believed to be synthesized in the interlocular septum of chili peppers and depends on the gene AT3, which resides at the pun1 locus, and which encodes a putative acyltransferase. Biosynthesis of the capsaicinoids occurs in the glands of the pepper fruit where capsaicin synthase condenses vanillylamine from the phenylpropanoid pathway with an acyl-CoA moiety produced by the branched-chain fatty acid pathway. Capsaicin is the most abundant capsaicinoid found in the genus Capsicum, but at least ten other capsaicinoid variants exist. Phenylalanine supplies the precursor to the phenylpropanoid pathway while leucine or valine provide the precursor for the branched-chain fatty acid pathway. To produce capsaicin, 8-methyl-6-nonenoyl-CoA is produced by the branched-chain fatty acid pathway and condensed with vanillylamine. Other capsaicinoids are produced by the condensation of vanillylamine with various acyl-CoA products from the branched-chain fatty acid pathway, which is capable of producing a variety of acyl-CoA moieties of different chain length and degrees of unsaturation. All condensation reactions between the products of the phenylpropanoid and branched-chain fatty acid pathway are mediated by capsaicin synthase to produce the final capsaicinoid product.
=== Logo === The design was created by McLean Design, a California-based strategic branding firm. The logo is composed of a vibrant green "M", composed of three lines on a field of black. The "M" is stylized in such a way as to imply that it is formed by the claws of a monster ripping through the can.
BASF's Functional Solutions segment consists of the Catalysts, Construction Chemicals and Coatings divisions. These divisions develop customer-specific products, in particular for the automotive and construction industries.

Researchers have cultured it in order to understand the biology of the eukaryotic cell and ultimately human biology in great detail. Other species of yeasts, such as Candida albicans, are opportunistic pathogens and can cause infections in humans. Yeasts have recently been used to generate electricity in microbial fuel cells and to produce ethanol for the biofuel industry. Yeasts do not form a single taxonomic or phylogenetic grouping. The term "yeast" is often taken as a synonym for Saccharomyces cerevisiae, but the phylogenetic diversity of yeasts is shown by their placement in two separate phyla: the Ascomycota and the Basidiomycota. The budding yeasts, or "true yeasts", are classified in the order Saccharomycetales, within the phylum Ascomycota.
=== Relationships and marriages === While in her late teens, Kidder became pregnant via a boyfriend, who arranged for her to undergo an illegal abortion. While filming 92 in the Shade, Kidder became romantically involved with Thomas McGuane, and in March 1975, relocated with him to Livingston, Montana. She subsequently became pregnant and gave birth to their only child, a daughter, on October 28, 1975. Kidder and McGuane married on August 2, 1976, but the marriage ended in divorce on July 21, 1977. During the marriage, Kidder stated that her self-esteem had faltered significantly, and she found maintaining a career in film to be difficult while residing in Montana. On August 25, 1979, she married actor John Heard, but the couple separated only six days into their marriage. Their divorce was finalized on December 26, 1980. Kidder was romantically linked to Canadian prime minister Pierre Trudeau in the early 1980s. Kidder married in 1983 French filmmaker Philippe de Broca, who had directed her in Louisiana. Her marriage to de Broca lasted one year, ending in divorce in 1984. Kidder later characterized the marriage as "impulsive, I'm afraid. Not a little irresponsible. We just weren't meant to be married to each other."
Viktor Antonov (Bulgarian: Виктор Антонов; 5 February 1972 – 7 February 2025) was a Bulgarian artist, video game designer, writer, and worldbuilder who worked on numerous first-person shooter (FPS) games. In 2017, Blake Hester wrote for Vice that Antonov "has created disturbing, memorable, and unique worlds" which "conjure images of cyberpunk metropolises and grim London alleyways." Antonov's first game credits were at Xatrix Entertainment, for Redneck Rampage (1997), Redneck Deer Huntin' (1998), Redneck Rampage Rides Again (1998), and Kingpin: Life of Crime (1999). At Valve, he worked on Counter-Strike: Source (2004), Half-Life 2 (2004), and Half-Life 2: Lost Coast (2005). As art director of Half-Life 2, he principally designed its dystopian setting of City 17: a decaying Eastern European city occupied by the Combine alien empire, featuring Soviet-era Brutalist buildings and alien structures similar to panopticons which constitute the Combine's surveillance state. Antonov was temporarily art director of Valve's Team Fortress 2 (2007). While developing Dark Messiah of Might and Magic (2006) and Dishonored (2012) at Arkane Studios, Antonov worked on films and wrote a graphic novel. He principally designed Dishonored's setting of Dunwall, a labyrinthine dystopian city with steampunk Victorian and Gothic buildings; it and City 17 are often considered some of the best worlds ever made for video games.

Male mammals typically have rudimentary mammary glands and nipples, with a few exceptions: male mice do not have nipples, male marsupials do not have mammary glands, and male horses lack nipples. The male dayak fruit bat has lactating mammary glands. Male lactation occurs infrequently in some species. Mammary glands are true protein factories, and several labs have constructed transgenic animals, mainly goats and cows, to produce proteins for pharmaceutical use. Complex glycoproteins such as monoclonal antibodies or antithrombin cannot be produced by genetically engineered bacteria, and the production in live mammals is much cheaper than the use of mammalian cell cultures.
=== Laboratory features === McLeod syndrome is one of only a few disorders in which acanthocytes may be found on the peripheral blood smear. Blood chemistry testing may show increased lactate dehydrogenase (LDH) indicative of hemolytic anemia or elevated creatine kinase when the patient is affected by myopathy (skeletal muscle damage).
Smoking has become less popular, but is still a large public health problem globally. Worldwide, smoking rates fell from 41% in 1980 to 31% in 2012, although the actual number of smokers increased because of population growth. In 2017, 5.4 trillion cigarettes were produced globally, and were smoked by almost 1 billion people. Smoking rates have leveled off or declined in most countries, but are increasing in some low- and middle-income countries. The significant reductions in smoking rates in the United States, United Kingdom, Australia, Brazil, and other countries that implemented strong tobacco control programs have been offset by increasing consumption in low income countries, especially China. The Chinese market now consumes more cigarettes than all other low- and middle-income countries combined. Other regions are increasingly playing larger roles in the growing global smoking epidemic. The WHO Eastern Mediterranean Region (EMRO) now has the highest growth rate in the cigarette market, with more than a one-third increase in cigarette consumption since 2000. Due to its recent dynamic economic development and continued population growth, Africa presents the greatest risk in terms of future growth in tobacco use. Within countries, patterns of cigarette consumption also can vary widely. For example, in many of the countries where few women smoke, smoking rates are often high in males (e.g., in Asia). By contrast, in most developed countries, female smoking rates are typically only a few percentage points below those of males.

It binds and activates both the GIP and GLP-1 receptors, making it a dual incretin receptor agonist. Single-receptor GLP-1 agonists act on one target only. The dual profile is the defining pharmacological feature of the molecule.
A fatty diacid side chain promotes reversible binding to serum albumin. This association slows renal clearance and protects the peptide from rapid enzymatic degradation. The result is a prolonged circulation time that supports weekly administration.
The first regulatory approval, for type 2 diabetes, was granted in the United States in 2022. An additional approval for chronic weight management followed in 2023. Availability and approved indications vary by country and are set by each national regulator.
It activates both GIP and GLP-1 receptors. This dual action differentiates it from selective GLP-1 agonists.
